To induce diabetes mellitus type I in rats, streptozotocin (链脲佐菌素STZ, 1% w/v, dissolved in citrate buffer, pH = 4.5) was intraperitoneally injected into SD rats (70 mg/kg). After 3 days, the rats were anesthetized by intraperitoneal injection of 10% (w/v) chloral hydrate（水合三氯乙醛）. Their backs were shaved, and a rounded full-thickness cutaneous wound area with the diameter of 1 cm was created.

Rats with blood glucose levels >16.7 mmol/L were defined as diabetic and maintained for another 4 weeks prior to wound induction. For the creation of surgical excisional wounds, STZ-induced diabetic rats were anesthetized with 1% sodium pentobarbital solution (40 mg/kg body weight, intraperitoneally). The dorsal hairs of the diabetic rats were subsequently shaved, and full-thickness excision (15 mm in diameter) was created on the dorsal surface.
