·492· 实验与检验医学 2018 年 8 月第 36 卷第 4 期 Experimental and Laboratory Medicine, Aug.2018,Vol.36,No.4 ·实验研究· 胶乳增强免疫比浊法测定 KL- 6 的方法学评价 吕某某，刘某某，王某某，应某某，叶某某 (XX大学第二附属医院检验科，江西 XX 330006) 摘要: 目的 对胶乳增强免疫比浊法（LEIA）检测唾液酸化糖链抗原(KL-6)进行方法学评价，判断其能否满足临床要求。 方法 在贝克曼库尔特 AU5421 全自动生化分析仪上利用 LEIA 法检测血清 KL-6，参考美国临床实验室标准化协会(CLSI)的 标准化评价方案评价 LEIA 法检测血清 KL-6 的精密度、回收率、线性范围、抗干扰性、稳定性等指标。 结果 LEIA 法检测血 清 KL-6 在（50～5000）U/ml 范围内线性良好（y＝0.9923 x-0.0776，r2＝0.9981），线性范围内偏差＜10%；高、低浓度样本批内不精 密度评价变异系数 CV 分别为 2.60％、2.63％，日间不精密度评价变异系数 CV 分别为 3.13％、4.07％；平 均 回 收 率 为 100.4%； 总 胆 红 素≤200mg/l、血 红 蛋 白≤6g/L、脂 肪 乳≤5%，对 该 法 无 明 显 干 扰 ；试 剂 在 仪 器 内(2～8)℃的 条 件 下 放 置 ，至 少 可 保 存 稳 定 29d；与化学发光法比较，回归方程为 y=0.9581 x+14.3206，r2=0.9989，两法高度相关（P＜0.01）。 结论 在全自动生化分析仪 上利用 LEIA 法测定 KL-6 操作简便、快速，检测结果准确可靠、重复好、线形范围较宽，与化学发光法比较有良好的相关性， 符合临床实验室要求。 关键词：唾液酸化糖链抗原 KL-6；胶乳增强免疫比浊法；方法学评价 中图分类号：R446.6 文献标识码：A 文章编号：1674-1129(2018)04-0492-04 DOI：10.3969/j.issn.1674-1129.2018.04.009 Evaluation of the latex -enhanced immunoturbidimetric assay in the measurement of the KL -6 LV Jiaofeng，LIU Jing， WANG Xiaozhong，YING Houqun，YE Shuang. Abstract：Objective To evaluate the latex-enhanced immunoturbidimetric assay (LEIA） for the detection of serum Kerbs von den Lungen-6 antigen (KL-6) and determine whether it can meet the clinical requirements. Methods Beckman Coulter AU5421 automatic biochemical analyzer was used to evaluate the analytical performance of LEIA for testing KL-6 according to the Clinical and Laboratory Standards Institute (CLSI) standardization evaluation protocal. The precision，recovery rate，linear range，anti-interference and stability were assessed. Results The linear range was from (50～5000)U/ml (y＝0.9923 x-0.0776，r2＝0.9981)，and the deviation within the linear range was less than 10%. The within-run coefficients of variation (CV) for high and low levels of KL-6 were 2.60% and 2.63%，respectively. The inter-day CV were 3.13% and 4.07%，respectively. The average recovery rate was 100. 4%. The serum samples containing total bilirubin ≤200mg/l，hemoglobin ≤6g/l and fat emulsion Injection ≤5% did not affect the result of KL-6. The reagent could be stable for 29d in the instrument under the condition of (2～8)℃.
